个人情况介绍
梁斌,博士,研究员(二级),博士生导师。2010年入选中国科学院“百人计划”,2014年入选“云南省高端科技人才”。从事“脂滴生物学与脂肪肝发病机理”研究。中国生物物理学会理事、代谢生物学分会理事,国际脂代谢领域知名期刊Journal of Lipid Research编委、Biophysics Reports 编委和青年编委主任。主持科技部国家重点研发计划重点专项(课题组长),国家基金委-云南省联合基金(2项)、重大研究计划、面上项目(3项),中科院先导专项B类项目。以通讯作者或第一作者在Cell Metabolism、Journal of Cell Biology、Nature Communications、Cell Reports等国际著名期刊发表SCI论文50多篇。获2020-2021爱思唯尔(Elsevier)中国金色开放获取高下载论文作者。

研究方向
1. 脂滴生物学
脂滴是重要的细胞器,存在于所有真核生物和部分原核生物。脂滴与其它细胞器(如内质网、线粒体、细胞核等)互作,响应胞内外环境变化,动态调节细胞的脂质代谢和能量平衡,参与生物体的生长、发育、生殖、衰老、抗逆境等。脂滴在脂肪组织和非脂肪组织(如肝脏)中大量沉积,导致肥胖、脂肪肝、糖尿病等代谢性疾病。我们利用秀丽线虫和小鼠等模式生物,重点研究:1)脂滴形成、增大、融合和分解的分子机理;2)脂滴蛋白靶向脂滴的分子机理;3)不同生理或病理条件下脂滴与其它细胞器互作维持细胞和机体脂质代谢和能量稳态的机理;进而揭示脂代谢紊乱相关疾病如脂肪肝的发病机理,寻找新的疾病标志物和治疗靶标。
2. 脂肪肝发病机理及治疗
肝脏是重要的代谢器官。临床上,当肝脏中多于5%的肝细胞出现脂滴积累时,即为肝脏脂肪变性。随着肝脏中脂肪过多的积累,脂毒性、胰岛素抵抗等多种因素作用下,出现小叶炎症和肝细胞损伤等,肝脏脂肪变性发展为脂肪性肝炎;如果脂肪性肝炎中出现胶原沉积则发展形成肝脏纤维化,继续恶化则导致肝硬化和肝癌。
我们利用脂肪肝细胞和动物模型,通过转录组、蛋白质组、代谢组等,重点开展脂质代谢和脂滴在脂肪肝中的作用机理研究,寻找新的疾病标志物和治疗靶标,肝脏疾病治疗药物的筛选、评价和作用机理研究。我们新发现一个脂肪肝致病因子——钙结合蛋白S100A11,其表达和脂肪肝的发病进展正相关,S100A11过表达导致脂肪肝和肝纤维化。目前正在研究S100A11在肝脏及其它代谢性疾病中的作用机理。
